Be at the entrance when they open, start with one of the most popular rides like Harry Potter ride, then venture down to the lower lot before going through the upper lot. We were able to do almost everything this way - only missing one ride by the time they closed. We saw all the shows too.

Universal is fun, but the lines for every ride can exceed 60-100 minutes on holiday weekends, spring/winter break, and all summer. I highly recommend purchasing the Express Pass during those times or you won't get to see more than 4-5 rides a day.
